I just recently purchased a 2006 Honda Shadow Spirit 750 that was not running well. Only one cylinder is firing. I have spark (all 4 spark plugs replaced), compression, and air. The forward cylinder is not receiving fuel. The fuel lines are clear. The fuel pump is good. Not sure what to do.

The bike has Vance and Hines pipes, and a K&N air filter.

Thats a two 34mm CV diaphram-type carb right? I would drop the float chamber and see if the float valve is OK.

You can drain the carb bowls to see if 1 or 2 ounces runs out of each.
If you have it running and spray carb cleaner into that carb will it pick and run strong?
If so the pilot jet is most likely clogged.

Yes. If I use carb cleaner, it runs with both cylinders.

So it is starving for fuel on that cylinder, probably the pilot jet. Time to take the carbs out.
But have you looked for leaks at the boots?
